abstract

the intracellular parasite neospora caninumis prevalent in several countries and is increasingly recognized as an important cause of abortion and stillbirth in cattle. for characterizeing the tachyzoite antigens of neospora caninumin aborted cows, sera were obtained from 116 cows which were aborted in the third semester of the pregnancy period and had antibodies to neospora caninumin elisa. to obtain the protein content of neospora, purified tachyzoites were lysed, electrophoretically separated and blotted to nitrocelloluse membrane for immunostaining. minimum 9 and maximum 13 protein bands ranging from 10 to 90 kda were observed after immunostaining. it seems that, in almost all of the cows, two protein bands with a molecular weight of 45 and 41 kda, have a prominent reaction in western blotting. according to our findings, these two protein bands are the most important antigens observed after western blotting, in seropositive aborted cows.

Temperature-dependent expression of flagella of Listeria monocytogenes studied by electron microscopy, SDS-PAGE and western blotting.

Washed cells of Listeria monocytogenes serotype 4b, grown in broth culture at 20 degrees C and at 37 degrees C, were examined by electron microscopy for the presence of flagella. Many flagella were seen in cells grown at 20 degrees C, whereas at 37 degrees C very few were expressed. Flagella sheared from the cell surface were partially purified by differential centrifugation. Using SDS-PAGE and...

SDS-PAGE and Western blot of urinary proteins in dogs with leishmaniasis.

Canine leishmaniasis is an endemic disease in the Mediterranean area caused by the protozoan Leishmania infantum, which usually produces renal failure. Sodium dodecyl sulphate polyacrylamide gel electrophoresis and Western blot using antibodies to IgG and IgA from dogs were carried out in the urine of 22 dogs with leishmaniasis diagnosed by ELISA and confirmed by PCR, and 20 healthy dogs. The r...

On-chip Western Blotting: In-situ Renaturation of Sds-protein Complexes Unifies Sodium Dodecyl Sulfate (sds) Sizing and Blotting in One Microdevice

Western blotting readily identifies specific proteins amidst complex biological backgrounds[1, 2]. Nevertheless, immunoblotting suffers from tremendous labor-intensive and time-intensive requirements [3]. The slab-gel assays require 1-2 days for completion with multiple hands-on “blotting” steps and yield semi-quantitative information. Recently, our group has introduced new approaches for compl...
